I'm assuming you're reffering to using these roms with Wii64 as it is the only app that can run N64 roms.
Anyways, while Wii64 can run roms off of a dvd you should note that the emulator is far from complete. Even with the latest version from the svn, you're still going to experience crashes, code dumps, and the inability of some roms (goldeneye 007 for instance) to work.
Now to answer your question, if you've successfully burned roms to a dvd and successfully ran them on other emulators (snes emu, nes emu, etc) you could probably burn n64 roms under the same settings. Always remember to use a slow burn speed (no matter what you're burning) and remember to use dvd-r discs or dvd+r discs withappropriate adjustments on your burner program.
